Restriction digests of the clone give the following sizes (kb): EcoRI--2.7, 0.34; HindIII--3.1; BamHI--3.1; PstI--3.1; PvuII--2.3, 0.68. CYP2A (pMP12) and CYP2B (pMP17) hybridize to the same MluI (350 kb), NotI (520 kb), and SstII (350 kb) fragments, but to different BssHII, NaeI, NarI, SfiI, and SmaI fragments. Although this is a cDNA clone, the insert corresponds to intron 5 which appears to have been retained as the result of an alternate splicing event in which exon 8 is missing and intron 5 is present. Parts of this probe contain Alu repeat elements. BglII digestions reveal polymorphism with one allele at 16.1 kb and invariant bands at 20.7 kb and 10.3 kb. MspI digestions also reveals polymorphism with only one allele at 2.9 kb (0.51) seen clearly, and invariant bands at 3.5 kb, 2.2 kb, and 1.0 kb. Enzyme(s) not detecting polymorphism: ApaI, EcoRI, EcoRV, HaeIII, HindIII, HinfI, PstI, PvuII, RsaI, SacI, TaqI and XmnI. |
