Restriction digests of the clone give the following sizes (kb): BamHI--4.9, 2.9; BstEII--7.6; SstI--7.0, 0.4; NotI--7.6; EcoRI--5.4, 2.6. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-48 of processed human factor VII (Gla domain), with mutations A1Y, A3S, F4G, 5K, K32E, D33N, A34T, K38T, L39E, replace the corresponding region of factor IX. 5K indicates the insertion of Lys. |
